IHC image of PACO60825 diluted at 1:400 and staining in paraffin-embedded human pancreatic tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. |
Background: | Binds differentially to the SH3 domains of certain proteins of signal transduction pathways. Binds to phosphatidylinositols; linking the hemopoietic tyrosine kinase fes to the cytoplasmic membrane in a phosphorylation dependent mechanism. |
Synonyms: | SH3 domain-binding protein 2 (3BP-2), SH3BP2, 3BP2 |
UniProt Protein Function: | SH3BP2: an adaptor protein that binds differentially to the SH3 domains of certain proteins. Links the hemopoietic tyrosine kinase Fes to the cytoplasmic membrane in a phosphorylation dependent manner. Regulates FC-epsilon RI -mediated degranulation in rat basophilic leukemia RBL-2H3 cells. Expressed in a variety of tissues including lung, liver, skeletal muscle, kidney and pancreas. Defects in 3BP2 are the cause of cherubism, an autosomal dominant inherited syndrome. Two differentially spliced isoforms have been described. |
UniProt Protein Details: | Protein type:Adaptor/scaffold Chromosomal Location of Human Ortholog: 4p16.3 Molecular Function:phosphotyrosine binding; protein binding; SH3/SH2 adaptor activity Biological Process: signal transduction Disease: Cherubism |
NCBI Summary: | The protein encoded by this gene has an N-terminal pleckstrin homology (PH) domain, an SH3-binding proline-rich region, and a C-terminal SH2 domain. The protein binds to the SH3 domains of several proteins including the ABL1 and SYK protein tyrosine kinases , and functions as a cytoplasmic adaptor protein to positively regulate transcriptional activity in T, natural killer (NK), and basophilic cells. Mutations in this gene result in cherubism. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2009] |
