Western Blot. Positive WB detected in: 293 whole cell lysate. All lanes: CAMK2A antibody at 1:2000. Secondary. Goat polyclonal to rabbit IgG at 1/50000 dilution. Predicted band size: 55, 56 kDa. Observed band size: 55 kDa.
Immunofluorescence staining of Hela cells with PACO64183 at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
IHC image of PACO64183 diluted at 1:200 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. |
Background: | CaM-kinase II (CAMK2) is a prominent kinase in the central nervous system that may function in long-term potentiation and neurotransmitter release. Member of the NMDAR signaling complex in excitatory synapses it may regulate NMDAR-dependent potentiation of the AMPAR and synaptic plasticity (By similarity). Phosphorylates transcription factor FOXO3 on 'Ser-298'. Activates FOXO3 transcriptional activity (By similarity). |
Synonyms: | Calcium/calmodulin-dependent protein kinase type II subunit α (CaM kinase II subunit alpha) (CaMK-II subunit alpha) (EC 2.7.11.17), CAMK2A, CAMKA KIAA0968 |
UniProt Protein Function: | CAMK2A: a protein kinase of the CAMK2 family. A prominent kinase in the central nervous system that may function in long-term potentiation and neurotransmitter release. Member of the NMDAR signaling complex in excitatory synapses that may regulate NMDAR-dependent potentiation of the AMPAR and synaptic plasticity. The holoenzyme is composed of four different chains: alpha, beta, gamma, and delta. The different chains assemble into homo- or heteromultimeric holoenzymes composed of 8 to 12 subunits. May interact with BAALC, MPDZ, SYN1 and synGAP. 2 isoforms of the human protein are produced by alternative splicing. |

NCBI Summary: | The product of this gene belongs to the serine/threonine protein kinases family, and to the Ca(2+)/calmodulin-dependent protein kinases subfamily. Calcium signaling is crucial for several aspects of plasticity at glutamatergic synapses. This calcium calmodulin-dependent protein kinase is composed of four different chains: alpha, beta, gamma, and delta. The alpha chain encoded by this gene is required for hippocampal long-term potentiation (LTP) and spatial learning. In addition to its calcium-calmodulin (CaM)-dependent activity, this protein can undergo autophosphorylation, resulting in CaM-independent activity. Two transcript variants encoding distinct isoforms have been identified for this gene. [provided by RefSeq, Nov 2008] |
